Infections by the digenetic trematode, Ribeiroia ondatrae, cause severe limb malformations in many North American amphibians. Ribeiroia ondatrae also infects fishes as second intermediate hosts, but less is known about the pathology and immune responses initiated in infected fish, even though reports of infected fish date back to early 1900s. To this end, we experimentally exposed juvenile Bluegills Lepomis macrochirus to three doses of R. ondatrae cercariae and monitored the pathology, parasite infection success, and humoral responses over 648 h. All exposed fish became infected with metacercariae, and the average infection load increased with exposure dose. Histologically, infection was associated with acute hemorrhages in the lateral line and local dermis at 36 h, followed by progressive granulomatous inflammation that led to the destruction of encysted metacercariae. Correspondingly, over the course of 648 h we observed an 85% decline in average infection load among hosts, reflecting the host's clearance of the parasite. Infection was not associated with changes in fish growth or survival, but did correlate with leukocytosis and neutrophilia in circulating host blood. Understanding the physiological responses of R. ondatrae in Bluegill will help to clarify the ecological effects of this parasite and provide a foundation for subsequent comparisons into its effects on behavior, individual health, and population dynamics of Bluegill.

In this paper, the use of supercritical fluid extraction (SFE) and micellar electrokinetic capillary chromatography (MEKC) is proposed for the complete analysis of volatile nitrosamines in sausages. The extraction fluid used was CO2 and variables such as density, temperature of thimbles, extraction time, modifier, fluid flow, and kind of traps were investigated. Several experiments were carried out to obtain the most favorable conditions for analysis of volatile nitrosamines in sausages. The recoveries ranged from 21 to 82% for the five nitrosamines studied. The optimal condition of extraction was 0.2 g of sample fortified with 10 mg/kg, using dynamic extraction during 20 min and with adsorbent Florisil in the trap. Eight volatile products characterizing strawberry aroma, which is generated from the oxidative degradation of linoleic and linolenic acids by a lipoxygenase (LOX) pathway, were examined because of their antifungal activity against Colletotrichum acutatum, one of the causal agents of strawberry anthracnose. In this study, the effects of aldehydes, alcohols, and esters on mycelial growth and conidia development were evaluated. (E)-Hex-2-enal was found to be the best inhibitor of mycelial growth [MID (minimum inhibitory doses)=33.65 microL L(-1)] and of spore germination (MID=6.76 microL L(-1)), while hexyl acetate was the least effective of all volatile compounds tested (MID=6441.89 microL L(-1) for mycelial growth and MID=1351.35 microL L(-1) for spore germination). Furthermore, the antifungal activity of (E)-hex-2-enal on susceptibility of strawberry fruits to C. acutatum was also confirmed. The presence of these molecules in jars containing strawberry fruits inoculated with a suspension of spores inhibited the fungus growth and prevented the appearance of symptoms. Moreover, a study of the effects of (E)-hex-2-enal on conidial cells of C. acutatum was evaluated by transmission electron microscopy. Phenolic compounds are abundant vegetable secondary metabolites in the human diet. The ability of procyanidin oligomers and wine polyphenols to inhibit trypsin activity was studied using a versatile and reliable in vitro method. The hydrolysis of the chromogenic substrate N-benzoyl-d,l-arginine-p-nitroanilide (BApNA) by trypsin was followed by spectrophotometry in the presence and absence of condensed tannins and wine. A clear relationship between the degree of polymerization of procyanidins and enzymatic inhibition was observed. Trypsin activity inhibition was also detected in several types of wine. In general, the inhibition increased with the concentration of phenolic compounds in wines. These results may be relevant when considering these compounds as antinutritional factors, thereby contributing to a reduced absorption of nutrients. 相似文献
The effects of fruiting body maturity on chemical composition and antimicrobial activity of the wild mushrooms, Lactarius deliciosus and Lactarius piperatus, were evaluated. Immature and mature samples were analyzed for proximate constituents (moisture, fat, crude protein, ash, carbohydrates), nutritional value, and individual composition in fatty acids and sugars. Protein content, MUFA (monounsaturated fatty acids), and PUFA (polyunsaturated fatty acids) percentages increased with the fruiting body maturity stage, while carbohydrate and SFA (saturated fatty acids) content decreased. The maturity stage did not significantly affect the individual sugar profile. The antimicrobial activity of the mushrooms was screened against Gram positive and Gram negative bacteria and fungi, and correlated to the amounts of phenols, flavonoids, ascorbic acid, beta-carotene, and lycopene present in the immature and mature fruiting bodies. Mature fruiting bodies with mature spores presented lower antimicrobial activity, which was in agreement with the bioactive compound content found in those samples. 相似文献

Yield mapping technologies can help to increase the quantity and quality of agricultural production. Current systems only focus on the quantification of the harvest, but the quality has equal or greater importance in some perennial crops and impacts directly on the financial profitability. Therefore, a system was developed to quantify and relate the quality obtained in the classification line with the plants of the orchard and for decision-making. The system is comprised of hardware, which obtains the location of the harvester bag during harvesting and unloading at the unloading site, and software that processes the collected data. The cloud of real-time data contributed from the different collectors (bins) allows the construction of yield maps, considering the multi-stage harvesting system. Further, the system enables the creation of a detailed map of the plants and fruits harvested. As the harvest focuses on quality, it takes place in stages, depending on the ripening of the fruits. In addition to the yield maps, the system allows identification of the efficiency of each worker undertaking the harvest by the number of performed discharges and by the time spent. The system was developed in partnership with the Federal Technological University of Paraná and Embrapa Uva & Vinho and was tested in apple orchards in southern Brazil. Although the system was evaluated with only data from apple cultivation, monitoring the quality and quantifying other orchard fruits can positively impact the fruit sector.
The development and intensification of Nile tilapia farming systems has happened together with ever increased use of antibiotics and therapeutants. Research on the use of probiotics has come along as alternative to routine use of antibiotics in fish diets. The manipulation of the gut microbiota through dietary probiotic inclusion targeting improved growth rate and disease resistance has been investigated in many farmed fish species. However, mechanisms underlying modulation of fish immune response remains unclear. This review summarizes and discuss recent findings in the role of probiotics on the immunity of Nile tilapia Oreochromis niloticus addressing available information regarding immunological parameters such as phagocytic activity, lysozyme, respiratory burst activity, antioxidant enzymes, complement system as well as immune-related gene expression, in the aim to foster research efforts on effective strategies for the successful use of probiotics in the tilapia farming industry.
This study investigated the overwintering survival and infectivity of free-living gastrointestinal nematode (GIN) stages on pasture. The presence of GIN larvae was assessed on 3 sheep farms in Ontario with a reported history of clinical haemonchosis, by collecting monthly pasture samples over the winter months of 2009/2010. The infectivity of GIN larvae on spring pastures was evaluated using 16 tracer lambs. Air and soil temperature and moisture were recorded hourly. Free-living stages of Trichostrongylus spp. and Nematodirus spp. were isolated from herbage samples. Gastrointestinal nematodes were recovered from all tracer lambs on all farms; Teladorsagia sp. was the predominant species. Very low levels of Haemonchus contortus were recovered from 1 animal on 1 farm. The results suggest that Haemonchus larvae do not survive well on pasture, while Teladorsagia sp., Trichostrongylus spp. and Nematodirus spp. are able to overwinter on pasture in Ontario and are still infective for sheep in the spring. 相似文献
